Utilizada no tratamento da gota, da febre mediterrânea familiar e outros transtornos como amiloidose, síndrome de Behçet, pericardite, cirrose biliar primária e pioderma gangrenoso. Está sendo testada sua eficácia no tratamento da COVID-19 (Cumhur 2020). Administração oral em uma ou duas doses diárias. É excretado no leite materno em quantidade que difere segundo diversos trabalhos, desde clinicamente não significativa em uns (Ben-Chetrit 1996, Milunsky 1991), até importante em outros (Guillonneau 1995), mas não se observaram problemas em bebês que mamam cujas mães a tomavam, até durante meses (Duman 2019 e 2016, Herscovici 2015, Diav-Citrin 2010, Ben-Chetrit 1996, Guillonneau 1995, Milunsky 1991), e os níveis na urina desses bebês que mamam foram indetectáveis (Guillonneau 1995). Não administrar simultaneamente com macrolídios ou sucos de frutas (nem a mãe nem ao bebê que mama), especialmente de toranja, pois aumentam consideravelmente a toxicidade por diminuir sua excreção biliar (Finkelstein 2010, Goldbart 2000). Diversas sociedades médicas e consensos de especialistas consideram o uso desta medicação seguro durante a amamentação (Sammaritano 2020, Götestam 2016, Yanmaz 2014, Finkelstein 2010, Stankovic 2008, Ben-Chetrit 1998). Academia Americana de Pediatria: medicação usualmente compatível com a amamentação (AAP 2001). Lista de medicamentos essenciais OMS 2002: compatível com a amamentação (WHO 2002).
